DNA sequencing is the process of determining the sequence of nucleotides (A,T,G,C) in a piece of DNA.

Sanger sequencing is a method of DNA sequencing developed by Frederick Sanger and colleagues in 197. It was the most widely used sequencing method for approximately 40 years. Later on, higher volume Sanger sequencing has been replaced by "Next-Gen" sequencing methods, especially for large-scale, automated genome analyses. However, for smaller-scale projects, and for validation of Next-Gen results Sanger method remains in wide use.

In Sanger sequencing, the target DNA is copied many times, making fragments of different lengths. Fluorescent “chain terminator” nucleotides mark the ends of the fragments and allow the sequence to be determined whereas in the Next-generation sequencing techniques are newer, and large-scale approaches are there that increase the speed and reduce the cost of DNA sequencing.
