Answer #1

Answer 1

Option A is correct " no colonies on transformation plate".

Explanation

Negative control does not contain any plasmid that gives resistance to antibiotics present in the agar plate in that case all bacterial cells with no plasmid carrying antibiotics resistance gene will not survive. Thus, no colonies can be observed.

Answer 2

Option A is correct " no colonies on transformation plate".

Explanation

Because transformants only contains gene that gives resistance against ampicilin. No resistance is present against kanamycin so all the bacterial cells will die and no colonies can be observed in transformation plate.

Answer 3

Option C is correct i.e " many more colonies that than we would get with ampicilin".

Explanation

If no antibiotics or ampicilin will be present in agar plate then no resistance will be required by bacterias to grow. So, in that case both transformants as well as non-transformants will grow and results in more number of colonies.

Answer 4

Option B is correct i.e " to degrade the ampicilin".

Answer 5

Option D is correct i.e " a and b are both true".

Explanation

This recovery step is required for the cells to grow, come out of harsh conditions and can amplify the plasmid carry ing gene of interest and can also express the gene that gives resistance against antibiotics. Thus, recovery period gives cells the ability to survive in presence of antibiotics.
