INFORMATION

Quadrant streak isolate the colonies of different group of bacteria
gram stain differentiate gram +ve and gram -ve bacteria crystal violet stain; counterstain purple color(gram +ve)pink color(gram -ve) no stain differentiate bact. by their physical and chemical properties of cell wall by staining process.
motility agar differentiate between motile and non motile bacteria agar spreading throught out the medium present only the area of incubation non motile bacteria grow only in soft agar tube.
eosin methylene blue agar identify gram -ve bacteria that fermet lactose blend of two stain (eosin &methylene blue nucleated colonies with purple & black colorless colonies lactose fermentation lower the ph that increases dye absorpation
SIM

1.sulfide

2. indole

3. mobility

sodium thiosulphate and peptone including tryptophan

black color( + for sulfur reduction)

red color (+ for indole production) growth all over the tube ( motile)

no color change
brewers plate in gaspak cultivating anaerobic and microaerophilic bacteria brewer anaerobic agar with sodium thioglycollate show growth in absence of o2 no growth
fluid thioglycollate cultivation of areobic , anaerobic & microaerophillic organism sodium thioglycollate and agar aerobes(O2 conc highest) bact gathered on top of tube anaerobic( O2conc lowest) gathered in bottom microaerophilic gathered in upper part but not in top
citrate slant differentiating the gram + bacteria citrate bromothymol indicator turn green to blue color in ph > 7.6 no color change if ph < 6.9 bact. use citrate and form bicarbonate and carbonate which increases the ph.
carbohydrate test media classify bact. fermenter gas and acid production & non fermenter carbohydrate source

acid production (phenol red indicator turn into yellow)

gas production ( gas bubble in tube)

no change in color of phenol red indicator

no gas bubble form in test tube

phenylalanine to determine the ability of organism to deaminate tophenylalanine to phenylpyruvic acid phenylalanine green color formed no green color formed
urea agar differentiate enterobacterieceae

  • You are given a mixed culture of two organisms in a small tube of TSB.

You are given a mixed culture of two organisms in a small tube of TSB. These two organisms include a Gram-negative strain (your bacterial strain to identify) and a Gram-positive strain (a contaminant). Your first task is to isolate these organisms which will help you identify which Gram-negative strain you are working with. 1) You inoculate two streak plates on TSA media and MacConkey Agar using aseptic technique. For each medium, state whether the medium is selective, differential, or neither and...
