Ans 2- Each DNA molecule inside the cells contains two strands joined together by hydrogen bonds. Change in conditions can "denature" the DNA and cause these strands to separate. Adding strong bases, like NaOH, increases the pH, thus decreasing the hydrogen ion concentration of the solution and denaturing double-stranded DNA.

Ans 3- The helical structure of double-stranded DNA is destabilized by increasing temperature. Above a critical temperature (the melting temperature), the two strands in duplex DNA become fully separated.

Ans 4- Yes, the results would be affected if the DNA is chemically sheared. Shearing results in random breakpoints and so generates a smear of DNA on a gel.
