Answer #1

In a lacI- strain of E. coli, the lac operon will be expressed both in the presence or absence of lactose.

The expression will be constitutive because the transcription of lac structural genes will not be inhibited when lacI mutation prevents the lac repressor protein from binding to the operator.

consider a strain of E. coli that has a mutation in the gene encoding the IIA protein of the phosphotransferase system. The transport and phosphorylation of glucose is not affected by this mutation and the phosphate group is transferred to glucose as in the wildtype strain. Also, the interaction between the mutant IIA protein and Lacy is not affected by the mutation. However, the mutation does affect the IIA protein's interactions with adenylate cyclase and the phosphorylated form of the...

Allolactose acts as a signal molecule that binds to the LacI protein (repressor) and prevents it from binding to the Operator on the lac operon. But if the repressor is on the operator, no B-glactosidase can be made which means no allolactose can be made in the first place. So how is the repressor removed so allolactose can be made to stop the repressor from binding?

Three different strains of E. coli carry a mutation in the lac operon and/or laci gene. The production of B-galactosidase (+ present or - absent) is measured when lactose is present and absent from the medium. Assume the mutations involve only 1, 0, or Z. A merodiploid is constructed for each of the three strains. The plasmid carries a wild type lac operon and lacl gene. The production of functional B-galactosidase (+ present or - absent) is measured when lactose...

As a student project, you have isolated six new mutant strains of E. coli with altered behavior of the lactose operon. The strains are listed in the table below, together with their phenotypes (with regard to significant ?-galactosidase synthesis) in three specific situations. Columns 1 and 2 present the phenotypes of each mutant haploid strain. In column 1, the mutant is in an otherwise wild-type genome. In column 2, the genome also carries a nonsense-suppressor mutation (that is not present...
