​​​​​​Suppose, there is species X. With time, it has undergone evolution and evolve into X1 and X2 species. The original species has undergone speciation because anyone or more of its genes has changed in its sequence. Suppose gene Y, has undergone changes.

X has gene Y.

X1 has gene Y1.

X2 has gene Y2.

If Y has undergone speciation, than Y1 and Y2 will be seen at different places on a gel.

The two species with doubt that have undergone is creation will show differences in the sequence of the same gene. To confirm, we will done the following experiment,

We will isolate the DNA from both the organisms.

Amplify the specific gene with you have a doubt has undergone speciation using PCR.

Now run the amplified PCR product on gel electroforesis. If speciation have occurred, then bands for the genes for both the species will occur at different positions. It will indicate their different molecular weight which is due to the change in their sequence with the course of evolution.
