Phosphodiester bridges have negative charge which gives DNA molecules a net negative charge. Phosphodiester bridges have negative charge because the pka value of phosphate group is near zero which Keeps it negatively charged. The phosphate group contains 4 oxygen atoms bonded with phosphorus atom. One out of this four oxygen atom is negatively charged and thus phosphate group holds negative charge.
