The petri plate should be stored upside down which means the bottom should be on the upper side because when you store a petri plate, condensation takes place and if the plate is not inverted then water droplets will fall on the colonies and hinder the development. Evaporation of water from media can cause media dryness and this can affect microbial growth but when plates are stored in inverted position, this decreases the rate of water evaporation and results in better microbial growth.
