Answer #1

Derivatization of an elute is the introduction of groups into the elute which increases the sensitivity to UV absorption.

Performing triplicate analysis of samples after initiating the derivatization will lead to a number of peaks showing variation in wavelength of absorption.

As we initiate the derivatization, some of the molecules will get derivatized and others will remain as it is because it will take time for whole sample to get derivatized.

Due to this the peaks after the absorption of UV light will contain peaks of the derivatized sample and low resolution of underivatized sample and so there will be errors in the resolution of the samples and will be unable to detect the samples with precision.

And after doing its triplicate analysis we will get different peaks everytime corresponding to the degree of derivatization which will take place.
