You are studying the live biosynthetic pathway in bread mold that produces a protein necessary for life. Your research group has identified three genes believed to function in the live pathway and has created mutant lines for each: die-21, die-18, and die-1. You have been growing the mutant strains and a wild-type strain on deficient media that does not contain all four of the compounds involved in the live pathway: A, B, C, and D. To determine the order of the genes in the pathway, you add back the compounds one at a time and obtain the following results:

a) What is the order of the compounds in the pathway? (left to right = first compound to end compound)

  1. A – B – C – D
  2. C – A – D – B
  3. B – C – D – A
  4. B – D – A – C
  5. D – C – B – A
  6. C – B – A – D

b) What is the order of the genes/ enzymes in the pathway? (left to right = first gene to end gene)

  1. Die-21 – Die-18 – Die-1
  2. Die-21 – Die-1 – Die-18
  3. Die-18 – Die-1 – Die-21
  4. Die-18 – Die-21 – Die-1
  5. Die-1 – Die-18 – Die-21
  6. Die-1 – Die-21 – Die-18

Answer #1

Answer a) The order of the compounds in the pathway (left to right = first compound to end compound) is option D ( B- D- A-C) because Biosynthetic pathway is blocked by mutation which cancels the enzyme needed for growth.

Answer b) The order of the genes/ enzymes in the pathway (left to right = first gene to end gene) is option C ( Die-18Die-1 die 21) because Biosynthetic pathway is blocked by mutation which cancels the enzyme needed for growth.

    34. (6 points) Currently you are studying threonine synthesis in yeast. To find genes whose protein products are important for threonine synthesis, you perform a mutagenesis to find mutants that require a source of threonine to grow. You find a total of nine mutants, all of which have recessive phenotypes. You perform complementation tests with all of your mutants. Below are the results for your complementation tests. t cells that are wildtype for the genes in the adenine pathway are of growing on minimal media lacking adenine. In parallel experiments, we O treat a wildtype culture of yeast cells with one of three mutagens: nitrous acid (a deaminating agent), UV light, and the intercalating agent, acridine orange. We obtain several adenine deficient colonies [ade- with each of the treatments and one mutant from each treatment condition is chosen for further analysis. DNA sequence analysis reveals that each...

(2 pts) The YPD plates contain a limited amount of adenine, which causes induction of the pathway to synthesize more. Mutation of the genes required for the pathway can lead to blockage at an intermediate stage of the reaction. Accumulation of a certain intermediate leads to a red coloration of the yeast in your Aade2 mutant. The order of the genes involved in the pathway can...

    32. Study the signaling pathway illustrated below, which involves two different receptors. Assume that a protein is active only when it is activated, but not inhibited, by its upstream signaling molecules. Activation and inhibition are indicated by normal and blunt arrows, respectively. Assume that the transcription regulator can be activated by either the binding protein or the kinase that are immediately upstream of the transcription factor.
