Ninhydrin is a chemical compound which reacts with amines to produce a chromophoric compound. We can measure protease activity using ninhydrin as follows.
Ninhydrin reacts with products of protease enzyme, and leads to formation of colored compound. The intensity of color, tells about the activity level of protease enzyme.

Ninhydrin + Colorless product Colored compound (ninhydrin-product complex)
We measure absorbance at 620 nm because the nihydrin - product complex has maximum absorption at 620 nm.
At 550 nm, we would be detecting for cytochrome c .
