4. After separating the component that is present in the food color mixture, wh have to do some other experiment to confirm the green color. After separation via any of the separating technique (column, paper, chalk, HPLC chromatographic), you have to dissolve in the solvent and you can find the green color in naked eye ad if you perform the UV vis spectroscopic study then you will get an absorbance band around 550 regions. This band reveals the green color.

5. To separate a mixture containing four amino acids, we have to perform a column chromatography. We have to increase the eluent polarity gradually to get the four different amino acids separately.

6. To separate the amino acid via paper chromatography, we have to do it very carefully. We should not touch the paper with our hand, because our body is full of amino acid which will make contamination with the paper and other amino acid mixture. During the addition of ninhydrin, it will show the different spot that is coming from our hand.

But this careness is not required for the food green color separation, because it will not be contaminated.
