Calcitriol is produced in the cells of the proximal tubule of the nephron in the kidneys by the action of 25-hydroxy vitamin D3 1-alpha hydroxylase,and an enzyme which catalyses the hydroxylation of 25-hydroxy cholecalciferol in the 1-alpha position.

Calcitriol is used to treat and prevent low levels of calcium and bone disease in patient whose kidney or parathyroid glands are not working normally.

The liver and Kidney convert the calcidiol into calcitriol .Calcidiol undergoes hydroxylation in the mitochondria of kidney tissue and this reaction is activated by the renal 25-hydroxy vitamin D3-1-(alpha)--hydroxylase to produce calcitriol, the active form of Vitamin D3.
