In a case file, staining for respiratory secretions, yielding negative for bacteria and positive for fungi shows that it is fungal pathogenic organism, not bacteria. The fungal organism is Candida albicans. Candida albicans is a pathogenic yeast of the human gut. It is commensal organism and found in the mouth and gastrointestinal tract in 40-60% of human individuals. In immunocompromised individuals, it can be pathogenic. It results in overgrowth of fungus.

EMB is Eosin methylene blue dye whereas in Macconkey agar the selective components are crystal violet and bile salts. On EMB agar, lactose fermenting organism produces black colonies and nonlactose gives colorless or light pink colonies. While Macconkey agar produces red-pink colonies with lactose fermenting microorganism and colorless colonies with non-lactose fermenters.
