Option D, EMSA Electrophoretic Mobility Shift Assay, In this technique it uses the mobility (movement) of the DNA in electrophoretic gel to determine whether it is attached with protein or not.

As the protein molecules are bigger in size and once it is attached to DNA , it reduces the mobility of the DNA molecule compared to the DNA molecule without the protein molecule.

hence the DNA fragment with protein (low mobility) will be at the top and DNA without protein (High Mobilty) will move downwards in the electrophoretic gel.
