Answer #1

The answer is option C

Cyclobutane thymine dimer cannot be repaired by Base excision repair.

The cyclobutane thymine dimer is a DNA lesion produced in the skin and is major cause for skin cancer.

The cellular epidermis is rich in UV absorbing chromophores (nuclear DNA being the most important) and are highly vulnerable to photodamage.

The dimers form when the C5=C6 double bond splits after absorbing UVR and links the two dimers at C5 and C6 positions.

The photodamage of DNA is very common and most organisms have elaborate DNA repair mechanisms.

Nucleotide excision repair is the most important one in human skin. This uses various enzymes for recognition of DNA photolesions using the opposite strand as template.

Base excision repair repairs the damage to to non-distorting single base modification caused by oxidation. Thus, it cannot repair the Cyclobutane thymine dimer lesion which is a distorted lesion.

DNA photolyase uses blue light to repair the diner lesion, by splitting the cyclobutane into two normal bases.

Thus, the wrong statement is that the Cyclobutane thymine dimer can be repaired by Base excision repair(BER).

and Lab Exer n this experiment you will evaluate effect of UV light on bacterial growth. UV light affects growth by interfering with DNA, resulting in thymine dimer formation. The dimerization is confined within adjacent thymine residues on same strand (intrastrand ). These dimmer lesions have consequence on DNA functions including interference in DNA replication, transcription etc, so UV exposure of cells can lead to lethal effects for bacterial multiplications. This forms the basis for UV method to control bacterial...
