GAP protein is GTPase activating Protein which bind to G protein and help to deactivating the G protein by helping in hydrolysis of bound GTP and convert it to GDP. As a result G-protein became deactivated. If due to the mutation GAP cannot bind to activated G protein, then G protein remain in activated form (ATP bound form) and cannot be deactivated (ADP bound form) as bound GTP cannot be hydrolyzed to GDP by GAP protein. So the cell signalling which is mediated by G protein always remain turned on and the cell signalling mediated by G protein continues. This has deleterious effect in cell because the regulation that terminates G protein mediated signalling is lost.
