The FRET-based cAMP indicator includes two major components :
Changes in conformation, or distance between the interacting protein partners, influence the distance or orientation of the two fluorophores and thus the energy transfer efficiency. The real-time cAMP imaging is based on fluorescein-labeled PKA-C and rhodamine-labeled PKA-R subunits, which have been microinjected into cells. Within a resting cell, FRET occurs between the two fluorophores under low cAMP conditions, while an increase within cAMP concentration is associated with a decrease in FRET due to dissociation of PKA subunits.
