After designig primers to target a gene of interest, you recieve 33.4 nmoles of a forward primer from the company. You want to make a 100uM stock but you accidentally made a 1mM stock solution. How much water do you need to add to your 1 mM stock to make it 100 uM?

Molarity or molar concentration is the amount of solute (measured as mole) per unit volume of solution ( measured in litres).

Hence, Molarity = no. of moles of solute ÷ volume of solution in litre.

In the question, we have 33.4 n moles of forward primer. With this amount of the primer, to obtain a 100uM solution, we need 0.000334L of water (334 uL of water).

Since, Volume of solution(L) = no.of moles ÷ molarity

But, instead a solution of 1mM was obtained. Volume of water in 1mM solution of 33.4nmole primer is 0.0000334L (that is 33.4 uL)

Hence, the volume of water required to convert a 1nM solution of 33.4n moles of a primer into a 100uM solution is the difference in the calculated water requirement for 100uM solution and the actual water content in1mM solution.

That is, amount of water required = 334uL -33.4uL = 300.6uL
