A. Hanging drop technique is used for viewing microorganisms/Bacteria without any stain. It is usually done to see motility of bacteria. In this method we put a drop of broth (containing growth of bacteria) over a slide and visualize it under microscope. As no stains are used therefore the shape, motility (as organism remains viable) can be well visualized.

B. Handing drop technique doesn't use any stain and most of bacteria are colourless and have relative low contrast to surrounding water medium. This makes bacteria difficult to visualize. To overcome this problem we adjust iris diaphragm, this enables us to increase contrast between bacteria and water. It also helps to focus light on small area. This helps in easy visualization of bacteria.
