Blood groups vary by the presence of carbohydrate moieties on the RBC membrane. The difference between A blood group and O blood group is the presence of an N-acetyl galactosamine group. So, if an enzyme can remove this molecule from A-blood group, it essentially becomes O-blood group.

So, the enzyme must be a glycosylase that can remove N-acetyl galactosamine.
