IS mutations change the repressor so that it can no longer bind the inducer. lac IS mutant is dominant, trans acting and uninducible.

these are known as superrepressed mutations. when studied in vitro, the mutant is polypeptides form tetramers that bind to lac operator DNA. they either donot bind inducer, or exhibit a very low affinity for inducer. the is mutations therefore modify the inducer binding siteof the lac repressor.

hence, the inducer cannot bind. no transcription and translation and thus no products.

there would be no products even in presence or absence of lactose.

Oc mutations are constitutive mutations. they act only in cis, i.e, they cause constitutive expression of structural genes present on the same chromosome.
