Answer

Pure RNA and duplex DNA hybridise and from DNA- RNA hybrid. This hybrid with large number of sequences and that are complementary. If the DNA and RNA are complementary then this means the percentage of G (guanine) and C (cytosine) in the RNA is always identical to the percentage of G and C in the DNA (duplex).

So if the RNA- DNA samples form hybrid then they must be complement to each other and this can be identified by calculating G and C percentage in both DNA and RNA (% of G and C must be identical).
