8. Anaphase promoting complex or "cyclosome" is an E3 ubiquitin ligase that plays very important role in regulating mitotic cell cycle, especially at the onset of anaphase and during mitotic exit. MAD1 (Mitotic arrest deficient 1), MAD2,BUBR1,BUB1, BUB3, and MPS1 are primary components of spindle checkpoint. Mad1-Mad2 heterodimers catalyzes a conformational change in another Mad2 at unattached kinetochores allowing binding and inhibition of cdc 20.

Hence, in presence of unattached kinetochores mad2-cdc20 interacts forming a diffusible MCC (Mitotic checkpoint complex) that will bind and inhibit APC/C. This will not allow cell cycle progression.
