Procedure 8.5 ESTIMATE THE TIME ELAPSED DURING THE PHASES OF MITOSIS Do all phases of the cell cycle require the same amount of time? The answer can be found by counting the number of cells in each stage of mitosis and interphase in a large number of cells in a tissue. An onion root tip cell takes approximately 24 hours to go through the entire cell cycle from one to two cells. You can calculate the amount of time spent in each phase of the cell cycle from the percent of cells in each stage. 1. Obtain a prepared slide of onion root tips or use the slide you used in Procedure 8.4. 2. Start with the scanning (5x) lens and then move to higher power lenses. Under high power (40X), focus on one field of vision on your prepared onion root tip slide. Identify and count the number of cells in each phase of mitosis and in interphase. Go down each row of cells. Tips: a. To avoid eyestrain, you may want to take turns doing this. For example, one student in the group can look for all the cells in interphase. Make a mark on a paper for each one you see. Then, without moving the slide, a second student can count the cells in prophase, and so on. b. If you can't focus at 40X, use 10X. c. To simplify the procedure, count the number of cells in each phase in one column of cells and multiply by the number of columns in your field of vision. 3. After you count the cells in one field of vision, move the stage, refocus, and repeat the process for one more field of vision.
