Question

You have isolated a tumorigenic cell out of a human brain biopsy from someone who has become a zombie. These brain cells refuse to die. You find that there is a protein you have named terminus that despite having the correct gene sequence, appears to be missing 2 domains of the protein that the non zombified form of terminus protein possesses. What would you suspect these differences result from? Design an experiment(s) to determine if your hypothesis is correct. Be sure to include what specific experiments you would do and what each experiment would tell us. Also, suggest a hypothesis as to how this zombie disease at the protein level could be spread by a bite from a zombie. 10 points. 5.

Answer #1

Answer :- To identify the reason why this terminus protein having less domain compare to the normal terminus protein or non zombie protein. For this study we had done following experiment to figure out the reason.

Experiment :-

We were using following technique to figure out the problem.

  • Reverse transcpitase PCR of the isolated mRNA.
  • Sequencing of DNA.

Experiment 01:-

We had to isolate the mRNA from a tumoregenic cell, so that we can form a c-DNA of mutated terminus protein.

For performing this experiment various protocols were persent according to our requirement.

Result:- c-DNA obtained .

Experiment 02:-

After obtaining the c-DNA we were go through the DNA sequencing process either by maxum gilbert or any other method through this sequencing we get the exact sequence of mutated DNA which is responsible for the expression of mutated terminus protein.

We will find out the bases responsible for the mutation by comparing with the given sequencing data of DNA of non mutated terminus protein.

Result :- Gene sequence have been identified which is responsible for mutation.

Hypothesis :- From the above experiment we hypothesized that this mutated terminus protein is responsible for the zombie formation because this protein passes from the infected person to the non infected during biting and cause the incomplete transcription of the RNA by unnecessary binding with the DNA and inhibit the process of transcription which results the formation of mutated terminus protein.

You are interested in studying the expression of Neurogenesis-2 (NG-2), a protein that may have a role in neurogenesis in the brain. You know that neurogenesis slows/stops as mice age, so you hypothesize that it will be highly expressed in young mice compared to old mice. (NG-2 is a fake protein used for this test question). Please note that you are being asked to analyze NG-2 at the PROTEIN level. Select appropriate techniques and technologies. (20 pts total). a) Describe...

please answer all that you can 1. You have genetically engineered green fluorescent protein (GFP) containing a KDEL sequence (GFP-KDEL). When GFP-KDEL is expressed in normal human fibroblasts and examined using fluorescence microscopy, the fluorescence appears diffuse across the cytoplasm. How would you explain this observations given that KDEL is supposed to be an ER-specific sorting sequence? A. This engineered GFP would not have a hydrophobic signal sequence to get it into the RER in the first place. B. The...
