Photorepair mechanism of DNA repair is a direct repair mechanism that brings the altered nucleotides back to their original and correct structure.

UV light is mutagenic and cause creation of pyrimidine dimers. This kind of DNA damage is corrected by the mechanism of photoreactivation repair of UV induced pyrimidine . This phenomenon was observed by Albert Kelner in 1949.

UV induce DNA damage which could be reversed if the cells are exposed briefly to light in blue range of visible spectrum. Process of photoreactivation repair is dependent on activity of protein enzyme called photolyase. This enzyme used energy captured from light to break the covalent bonds that link the pyrimidine in a dimer thus reversing the effect of UV radiation on DNA.

The enzymes must absorb a photon of light to cleave dimer.
