Three genes with recessive mutant phenotypes are located on the same chromosome in maize: waxy endosperm (wx), shrunken endosperm (sh), and virescent seedling (v). A plant with all three mutant phenotypes is crossed with a true-breeding, wild-type plant. Resulting F1 plants are then backcrossed to the parental mutant line to produce the following progeny:

Waxy Shrunken Virescent Number
wx sh + 89
+ + v 92
+ + + 1482
wx sh v 1475
+ sh + 480
wx + v 477
wx + + 290
+ sh v 282

Draw a chromosome map with the correct distances (rounded to the nearest cM) noted between

adjacent pairs of genes.

Here, wx wx sh sh v v(recessive mutant for all 3)..............X..............................wx+wx+ sh+sh+ v+v+(wild type)

Gametes ....wx sh v....................................................................................................wx+ sh+ v+

F1 ....................................wx+wx sh+sh v+v

Now the F1 progeny is crossed with recessive parent (test-cross)

Hence, wx+wx sh+sh v+v..........................X........................................................................................wx wx sh sh v v

Gametes: wx+ sh+v+,wx+shv,wx+sh+v,wx+shv+,wx sh v, wx sh+v+, wx sh+v, wx sh v+.....................wx sh v

F1 : wx+wx sh+sh v+v =(+++)= 1482

wx wx sh sh v v = (wx sh v) = 1475

wx+ wx sh sh v v= ( + sh v) = 282

wx+ wx sh+ sh v v= ( + + v) = 92

wx+ wx sh sh v+v = (+ sh +) = 480

wx wx sh+ sh v+ v= (wx + +)= 290

wx wx sh+ sh v v= ( wx + v) = 477

wx wx sh sh v+v = ( wx sh +)= 89

·         Largest number of offsprings: parental genotype of the heterozygous F1 plant =

wx+wx sh+sh v+v =(+++)= 1482 & wx wx sh sh v v = (wx sh v) = 1475

·         Smallest number of offsprings: double cross-over leading to these genotypes =

wx wx sh sh v+v = ( wx sh +)= 89 & wx+ wx sh+ sh v v= ( + + v) = 92

ORDER of gene: wx.v.sh (double crossin gover shown in diagram)

wx+ v+ sh+ = 1482

wx v sh = 1475

wx+ v sh = 282

wx v+ sh+ = 290

wx+ v+ sh = 480

wx v sh+ = 477

wx+ v sh+ = 92

wx v+ sh = 89

Now, Distance between wx & v = ( Single crossing over between wx & v + Double crossing over )/ Total =

((282+290)+ (89+92))/ 4667 = 0.161 or 16.1 cM

Now, Distance between v & sh = ( Single crossing over between v & sh + Double crossing over )/ Total =

((477+480)+(89+92))/4667 = 0.244 or 24.4 cM

Distance between wx & sh = ( Single crossing over between wx & sh + 2 X Double crossing over )/ Total =

((282+290+477+480)+ 2 X (89+92))/4667 = 0.405 or 40.5cM

Hence the chromosomal map is :

Patrick was hired by Dr. Ryan to work on a maize genome project. His research assignment was to map three linked genes which can cause brown midrib (bm), virescent seedling (v) and purple aleurone (pr) phenotypes respectively in maize. All the above mentioned alleles are recessive ones compared with the dominant wild type phenotype (designated with +). Dr. Ryan gave Patrick a triple heterozygote strain which has both the recessive mutant and dominant wild type alleles. Patrick was asked to...

Genetic Linkage The six genes listed below are all located on Chromosome 2 of Drosophila melanogaster. Your goal is to construct a genetic map of Chromosome 2. That is, determine the order of these genes along chromosome 2 and the map distance in centimorgans between each gene. To complete this task, you will be given the results of a variety of two-point test crosses involving these genes. For each test cross you may assume that the female is heterozygous and...

Researchers investigating chromosome 9 of com, Zea mays, found four mutations. Interested in finding the relative locations of the four mutations, they bred two different triple mutant strains. One of the strains displayed phenotypes of male sterility (ms), white endosperm cap (wec), and brown midrib (brm). The other strain displayed the phenotypes of male sterility, white endosperm cap, and brittle stalk (bst). They crossed both mutant strains with a true breeding wild type strain to create two F, hybrids. They...

    5. Deletions can be used to map genes along a chromosome. In order to do this a series of crosses in which one parent is homozygous for a mutant allele is crossed with the other parent that is homozygous for a partial deletion of the region. The progeny are scored to determine whether they have the mutant phenotype ("m") or the wild-type phenotype ("+"). If the mutant allele falls within the deletion it will have the "m" phenotype, if it...

In the tomato, three genes are linked on the same chromosome. Tall is dominant to dwarf, fruit skin that is smooth is dominant to skin that is peachy, and fruit with a normal tomato shape is dominant to oblate shape. A plant that is true-breeding for dominant traits was crossed to a dwarf plant with peachy, oblate fruit. The F1 plants were then test crossed to dwarf plants with peachy, oblate fruit. The following results are obtained. 200 tall, smooth,...
