4)Insert the sequence upstream of the coding sequence for a protein whose expression is easy to assay and then introduce that chimeric construct into cells and assay for protein expression.

Because if you put your sequence downstream of the coding sequence. It will not get expressed because your gene may have a stop codon. So putting upstream resolve this issue.
