Can you write about DNA quantification. Its purpose, and its advantages and disadvantages. What is the function of these molecules:

1.Ultrapure water
2. TE buffer
3. 70% EtOH

Answer:- DNA quantification is method use to measure concentration of nucleic acid (DNA, RNA). spectrophotometeric analysis is done to find out quantity of DNA, RNA in mixture by spectrophotometer. spectrophotometeric analysis is base on absorption of UV light by DNA when sample is exposed to UV light at 260nm wavelenght. More absorption of UV light higher the amount of DNA present in given mixture.

Optical density detected by = Log(intensity of incident light / Intensity of transmitted light)

then optical density multiply by conversion factor to obtain concentration in jg/ml . if if 10mm pathlengh is used then simply multiply optical density with 10 will give you concentration.

-Ultrapure water is mainly used for sterilise and cleaning purpose. as there is equilibrium between H+ and OH- ion which prevent any unwanted reaction with nucleic acid compare to normal water.

- TE buffer is used because it dissovle DNA , RNA and prevent degradation..

-70% EtOH precipitate DNA so we can get maximum amount of DNA.
