NRTIs act by inhibiting HIV-1 reverse transcriptase.

Common Side effects include nausea,vomiting,headache and abdominal discomfort.Other serious ones include Bone marrow suppression and Myopathies.

but the side effect which is problematic is Lactic acidosis.Once it happens its management is very difficult and it is a life threatening condition which requires immediate action. Bone marrow suppression can be reversed mostly by stopping the use of NRTIs and Myopathies are treated symptomatically but they do not require immediate treatment.
