The tRNA synthetase, load the specific amino acids to it's cognate tRNA. After charging with the amino acids the tRNA can be used for incorporation of amino acids to the growing polypeptide chain in the protein translation mechinary. During translation, ribosome read the mRNA from 5' to 3' direction and each triplet codon, code for incorporation of single amino acid. Just before the translation of mRNA, mRNA is been produced from the coding strand of DNA by the process called transcription.

All thses non-covalent and covalent (di-sulfide bond) bonds stabilizes the tertiary structure of the protein. Interfering any of these bonds will lead to distabization of tertiary structure.
