In your own words, what is FISH? Briefly describe how it works. How is FISH used to characterize chromosomal translocations associated with certain genetic disorders and specific types of cancers?

  • FISH is fluorescent in situ hybridization.
  • FISH is a Hybridization mapping technique by which DNA or RNA sequence in a gene may be determined.
  • RNA or DNA probe which is complementary and fluorescent tagged is specifically selected for the gene of interest.
  • The probes hybridize by formation of hydrogen bond with the desired sequence of gene of interest.
  • Thus, the tagged probe binds to the sequence to be detected and the location may be detected by observation through fluorescent microscope or color detection.
  • One of the important applications of FISH was used to analyze condition of Philadelphia chromosome.
  • The translocation results in abnormal chromosome with deletion –t(9,22)(q34,q11), resulting in neoplasm.
  • FISH may be used to detect the translocation by creating probes based on normal chromosome sequence and then detecting the sequence that translocated from long arm of chromosome 22 to short arm chromosome 9.
  • The probe that would bind to sequence in chromosome 22, would bind to the shifted sequence in chromosome 9 in the Philadelphia chromosome.
  • Thus, the translocation may be detected by the technique.
