i. Removal of primers by 5'-3' exonuclease activity of DNA polymerase or RNase H


ii. Replacement of primer with deoxyribonucleotides by 5'-3' polymerization activity of DNA polymerase


iii. Joining of two DNA fragments by DNA ligase
