Pi or inorganic phosphate is an essential substrate in a glycolytic reaction that catalyzed by a glyceraldehyde 3-phosphate dehydrogenase.

Glyceraldehyde-3-phosphate + NAD+ + Pi \rightleftharpoons 1,3-Bisphosphoglycerate + NADH

Primary phosphate is an essential substance to complete glycolytic pathway. Inorganic or primary phosphate, rather than ATP, provides the source of the phosphoryl group and it's absence may affect the formation of high energy phosphate bond in 1,3-Bisphosphoglycerate in glycolysis.
