Answer #1

1) Loop is sterilized after the initial inoculation in order to prevent the contamination

2) Pure culture: In pure culture only single type of the species is present

Mixed culture:  In mixed culture minimum two types of species are present in the culture

3) Bacterial colony:  Bacterial colony is the clump of bacteria growing together in the agar plate.

They are distinguished by :

i) shape

ii) surface

iii) Pigmentation

iv)Opacity

4) Petri Dish should not be kept open because It may get contaminated by the Microorganisms present in the air and the Agar gel will dry up soon

5) streaking method: it is a type of technique that gives us the isolated colonies and these isolated colonies were grown separately.

6) To grow individual species.

To observe locomotion of Protozoa in pond water, it is essential to make a "smear". Tor F 40. If a prepared smear is dried by heat it is likely to develop cell-shrinkage. Tor F 41. Smears must be thick to ensure that bacteria will be retained on the slide. Tor F 42. The size of bacteria on a NA Petri plate...

  • You are given a mixed culture of two organisms in a small tube of TSB.

You are given a mixed culture of two organisms in a small tube of TSB. These two organisms include a Gram-negative strain (your bacterial strain to identify) and a Gram-positive strain (a contaminant). Your first task is to isolate these organisms which will help you identify which Gram-negative strain you are working with. 1) You inoculate two streak plates on TSA media and MacConkey Agar using aseptic technique. For each medium, state whether the medium is selective, differential, or neither and...

please help with whatever possible. thank you so much in advance. Name One use of serial dilutions is to calculate the concentration of microorganisms. Since it would usually be challenging or even impossible to actually count the number of microorganisms in a sample, the sample is diluted and plated to get a reasonable number of colonies to count (usually between 25 to 250 colonies is the goal). Since each colony on an agar plate theoretically grew from a single microorganism,...
