Topoisomerase is an enzyme that breaks and rejoins the DNA strands to relive torsional strain in the DNA duplex due to folding or unwinding.
There are two types of topoisomerases.
1. Type I topoisomerase:
It cleaves and rejoins a single strand.
It does not require ATP hydrolysis for its function.
It is found only in eukaryotes
Three subclasses: IA, IB, and IC

2. Type II topoisomerase:
It cleaves and rejoins two strands.
It requires ATP hydrolysis for its function.
It is found in both prokaryotes and eukaryotes
Two subclasses: IIA and IIB

Negative supercoiling is naturally prevalent because it prepares the molecule for several DNA processes that require strand separation.
