Answer: Alzheimer's disease is caused due to accumulation of amyloid-β peptides in the brain. These peptides are produced by cleavage of amyloid precursor protein (APP) by beta-secretase and gamma-secretase. Beta secretase cleaves APP in to extracellular soluble fragment and a membrane bound C99 fragment. C99 fragment is cleaved by gamma secretase to produce β-amyloid. Drugs that block the activity of Beta secretase prevent the production of β-amyloid and is helpful in Alzheimer's disease. Hence, blocking the activity of Beta secretase would in theory be helpful in Alzheimer's disease because it would reduce the amount of amyloid produced.
