1) western blot is done to detect expression of a protein normally in cell or after the specific treatment to the cell . Appearance of a new band in western blot denote that treatment given to the cell has some effect on expression of that particular gene mainly enhancing expressikn of that particular gene.
As I dont know the previous scenario which you are talking about. So this is generalized answer.
2) increasing band density over time - increased expression of particular protein over time.
3) decrease band intensity over time - decreased expression of gene over time after treatment.
4) in EGF signaling as EGF bind to its receptor EGFR, there is activation of EGFR related pathway and there is increased expression of its downstream molecules which lead to increase band density of its downstream molecules
