1. Two signaling pathways process to the activation of NF-κB,namely canonical pathway and the non-canonical pathway.
a. CANNONICAL PATHWAY
NF-κB dimers are activated by IKK-( inhibitor of kappa B kinase) mediated phosphorylation of IκB( inhibitor of kappa B).
As IkB get phosphorylation it undergoes proteosomal degredation.
Degredation of IkB lead to formation of the active NF-κB (p50 & p65) transcription factor subunits to translocate to the nucleus and induce target gene expression.
b. NON CANNONICAL PATHWAY
IKKα complex phosphorylates p100 leading to the proteosomal degradation of p100 and liberation of the p52/RelB as active heterodimer.
