Option 1st is the correct answer. Michaelis-Menten equation represents a relation between the initial reaction rate (v0), the maximum reaction rate (Vmax), and the initial substrate concentration [S] through the Michaelis constant KM.

Michaelis-Menten equation is given by: V0 = Vmax [S]/ Km + [S]

Where V0 = initial velocity of reaction, Vmax = maximum velocity of reaction, [S] = substrate concentration and Km = Michaelis constant which represents the affinity of an enzyme towards the substrate and it is numerically equal to the substrate concentration at which the reaction rate is half of V max and indicates that the half of the enzyme molecules are bonded with given amount of substrate.
