Answer #1

pGLO plasmid contains the bla gene which is responsible for the ampicillin resistance this gene is always expressed.

the GFP is cloned under the arabinose promoter, it is expressed only in the presence of Arabinose, AraC gene codes for the regulatory protein, which controls the expression of the arabinose operon, AraC can inhibit the expression of the Arabinose operon in the absence of arabinose, so the GFP is expressed only in the presence of arabinose.

7) a) ampr ( or bla) ampicillin resistance due to the expression of beta-lactamase

8) b) GFP ( green fluorescent protein)

the enzymes for the arabinose metabolism are not present in the plasmid.

You want to use PGLO to produce red fluorescent protein (RFP) instead of green fluorescent protein (GFP). To do so you cut out the GFP gene from PGLO with two restriction enzymes, and replace it with the coding sequence for REP After you successfully created a modified version of the PGLO plasmid with the RFP in place of the GFP you transform this plasmid into E. coli and plate it onto LB plates containing both ampicillin and arabinose. After an...
