Answer #1

Comparative chart between the Western blot, Northern blot and Southern blot.

Method Applications Reagents Process
Western blot used to confirm a positive HIV diagnosis. methanol, glycine, dried milk, stains like Amido black, Alcian blue and Ponceau S. The process includes five steps namely transfer, blocking, primary antibody incubation, secondary antibody incubation and protein detection and analysis.
Southern blot used to detect specific DNA sequence from DNA sample. SDS, 6X SSC, 5X Denhardt's solution, yeast tRNA. The process involves transfer of electrophoresis separated DNA fragments to a filter membrane and subsequent fragment detection by probe hybridization.
Northern blot used to detect specific RNA molecules from a mixture of RNA. EDTA, SDS, Salmon sperm DNA, sodium heparin etc. The process includes separation of isolated RNA by gel electrophoresis, transferring to a membrane and detection of hybridization with a DNA or RNA probe.

    4. In future lectures we will describe a technique known as Northern blotting that can be used to detect RNA transcribed from a particular gene. Briefly, in this method a specific RNA is detected using a short segment of cloned DNA as a probe. The DNA probe, which is radioactive, is complementary to the RNA that the researcher wishes to detect. After the radioactive probe DNA basepairs to the RNA, the RNA is visualized as a dark (radioactive) band on...
