Conjugations are set up between E. coli of the genotypes indicated below. Bacteria are incubated for 10 minutes and then plated on minimal media with a concentration of azide (azi) sufficient to kill azi - sensitive E. coli . Predict how many Hfr, F+ and F - colonies (many, few, none) will appear on the azi plates and how they would come about. Note: the leu locus is very near the origin of transfer, whereas the azi locus is not.

A) Hfr azis leu+ x F- azir leu-

B) Hfr azis leu- x F- azir leu+

C) Hfr azis leu+ x F+ azir leu-

D) F+ azis leu+ x F- azir leu-

Could you please explain the reasoning behind why there would be that many colonies, I already have the answers. Also, could you explain what it means that the locus is near the origin of transfer in regards to how many colonies there would be? THANK YOU

2a. (4 pts) A potent mutagen was added to an E. coli culture growing in rich broth. Prior experiments established that this F cell line, designated WWU113, could not synthesize leucine. You are interested in measuring the rate of induced reverse mutation. You plan to grow a liquid culture of the leu strain and then plate various dilutions of the culture onto selective media. What media should you use to grow up the liquid culture? This will be Media A...

A generalized transducing phage is used to transfer genes from a thr(+)bio(+)thi(+) donor strain of E. coli to a thr(-)bio(-)thi(-) recipient strain. When transductants selected on a medium containing thiamine and threonine are tested for growth on other media, many more colonies are found on media supplemented only with threonine than on media supplemented only with thiamine. No transductants survive on minimal media. What is the correct order for these three genes? *I have seen multiple different answers for this,...
