2. The UAG codon is a stop codon because the stop codon is not recognized by any amino-acid charged tRNA , instead it is recognized by release factors which can bind and release the peptide formed during translation.

To change this aan altered tRNA needs to be generated which can recognize UAG codon and can bind to that codon. In that case even without changing the codon the translation can be altered.
