  • LAC operon of E. coli is an example of inducible gene expression system. Allolactose acts as inducer molecule in lac operon.
  • Lac i represents the repressor and is under a separate promoter and not under lac promoter control.
  • Lactose acts as inducer, which on binding to repressor, prevents it to bind to the operon and induces expression.
  • When there is low glucose concentration, high levels of cAMP are generated that binds to CAP, promoting transcription initiation of lac operon genes.
  • Lac i mutation is called super-suppressor, Lac is.
  • This makes the repressor insensitive to the inducer.
  • The repressor binds to the operator site.
  • If there is mutation in repressor gene, repressor is not produced and expression continues with or without inducer.
  • If there is mutation in the operator making it O-, the repressor is unable to bind, and transcription occurs even in absence of inducer. However, in merodiploid cell the operator is functional or O+. Thus, trans acting repressor can bind to the functional operator.
  • Mutant Oc operator is constitutive cis- dominant. The repressor cannot bind to this operator and thus, for cis-acting genes, there will be continuous transcription, even in absence of inducer.
