SUB CELLULAR FRACTIONATION: which involve separation of individual components from the cell by preserving the individual components.

  1. PURPOSE
  • Used to demonstrate the cellullar location of various biochemical processes.
  • provide an enriched source of protein
  • diagnosis of various diseases

main background of the cell fractionation involve the separation of the cell components by centrifugation cell fractionation and separating the organelles.

METHOD

The method involve following steps

  • breaking the cells- it involve the opening of cell membrane using the detergents and formation of cell homogenate it is also called lysing
  • Separation- it is done by the centrifugal force a super fast processing it will seprate the cellualar components according to mass, density, and shape.
  • collection- depend up on the liquid which is used for the centrifugation, usually used liquid providing the osmotic support such as sucrose or percoll only one concentration of the liquid is used it is called differential centrifugation

main advantages of the study is the this study is isolation of cell components and study of internal components diagnosis of diseases and it is reliable method. determination of mass , size of proteins can be determined by various analytical methods like mass spectroscopy , HPLC etc.

limitation include only separate components by size there for fractionation is difficult.
