Answer #1

During coinfection, the occurrence of lysis indicates that both the mutations are non-allelic i.e. they are located at different loci.
The non-occurrence of lysis indicates that both the mutations are allelic to each other i.e. they are located in the same locus.

A cistron is defined by non-complementation in the trans-configuration.

Number of genes mutated = Complementation groups = 3
i. 1 = 6
ii. 2 = 5 = 7
iii. 3 = 4 = 8

Strains 4 and 8 are defective in the same gene as that of strain 3.

As a student project, you have isolated six new mutant strains of E. coli with altered behavior of the lactose operon. The strains are listed in the table below, together with their phenotypes (with regard to significant ?-galactosidase synthesis) in three specific situations. Columns 1 and 2 present the phenotypes of each mutant haploid strain. In column 1, the mutant is in an otherwise wild-type genome. In column 2, the genome also carries a nonsense-suppressor mutation (that is not present...

After placing them in different mediums with different intermediates, she places some bacterial cells in a medium with Intermediate C and they survive and produce isoleucine. Please state which enzymes in this bacteria’s metabolic pathway are functional and which ones are not. -In a wild type strain of bacteria, she notices that when isoleucine accumulates its production starts slowing down, what type of regulation is this? Where...

Question 17. Saccharomyces cerevisiae (also called budding yeast or baker's yeast) normally makes LARGE colonies on the Petri dish. The fungus can exist as either a haploid or as a diploid. As a geneticist, you decide to isolate haploid mutants with altered size of colonies. You collect 6 such haploid mutants, which you arbitrarily name M1, M2, M3, M4, M5, and M6. To determine how many genes are represented by your mutants, you do a series of crosses resulting in...

Title: Development of an Adhesion Assay and Characterization of an Adhesion-Deficient Mutant of Pseudomonas fluorescens RESULTS Bacterial adhesion to sand columns. In the initial column assays with soil-and-sand mixtures, P. fluorescens PfO-1 and E. coli SLH25 had the highest and lowest adhesion values, respectively. Therefore, the sand column assay was optimized to yield the greatest difference in percent adhesion for these two strains. PfO-1 bacteria grown in minimal medium to the logarithmic phase were washed and suspended in either minimal...
