c. Would the pI of Glu-Pro-Lys-Asp be greater than or less than the pI of Glu-ProLys-Arg? Explain how to answer this question without going through the trouble of estimating both pI’s.
a) At pH 11, all groups except Arginine side chain exist in deprotonated state.
b) pI is the average of pKa values of group after and before neutral form. In this case it is average of pKa of Glu Amino group and Lys side chain.
pI = (9.67+10.79)/2 = 10.2
c) pI of Glu-Pro-Lys-Asp is smaller than pI of Glu-Pro-Lys-Arg because it contain acidic (Asp) Amino acid whereas later one contain basic Amino acid (Arg). Peptide having acidic Amino acid have low pI than that having basic Amino acid.
