Lab - Spectrophotometric determination of glucose content

Part 1

If the outside of your cuvette had not been properly cleaned, how would your result have been affected? would the amount of glucose be higher or lower? Explain.

Part 2

Why was it important to ensure that each sample was evenly heated for the same amount heated for the same amount of time? How would your results/graph been affected if the samples were exposed to different temperatures and heating times?

Solution:

Part 1) If cuvette is not properly cleaned, then some of light may be absorbed by the dirt and therefore absorbance reading increases on spectrophotometer.

Since, absorbance is directly proportional to concentration, hence the amount of glucose would be higher than actual amount of glucose.

Part 2) For the measurements iof absorbance in spectrophotometer, it js nessesory to evenly heating the samples for exact period of time because constant temperature stablize the sample. Thus, uneven heating causing both stablizing and unstablizing the samples, hence acuracy will be diminished.
